Abstrak

Abstract Background: Currently, there are various types of antidiabetic drugs available in the market. However, the use of synthetic drugs for the treatment of diabetes often causes problems, including side effects, resistance to therapy, and high treatment costs. SGLT-2 inhibitors, Pioglitazone, Metformin, Sulfonylurea, DPP-4 inhibitors (Gliptin), GLP-1 agonists, Acarbose, and other drugs are commonly used in diabetes pharmacotherapy. In addition to conventional oral antidiabetic drugs, natural substances such as probiotic Lactic Acid Bacteria (LAB) can be an alternative therapy for diabetes. Some types of probiotics have the ability to inhibit the activity of alphaglucosidase enzyme in intestinal microvilli, thereby reducing blood glucose levels. Objective: This study aimed to find suitable characteristics as probiotic candidates in dadih by screening and identifying molecular probiotics from curd as alpha-glucosidase inhibitors. Methods: This study used experimental method. Results: The results showed that isolate DDH8 had a growth percentage of 48% in the acid test, which showed significant bacterial growth, with a live growth rate of 46.595% and low bile salt tolerance. DDH 8 also showed moderate antibacterial activity, with average inhibition zone diameter of 10.5 mm against E. coli. Based on phenotypic and molecular identification using 16S rRNA, isolate DDH8 was identified as Lactobacillus fermentum, which has suitable characteristics as probiotic candidate. Conclusion: LAB isolates, identified as Lactobacillus fermentum through phenotypic and molecular identification based on 16S rRNA gene, showed potential as probiotics. LAB from curd was gram-positive, red in color, and rod-shaped. Screening results showed that this isolate was catalase-negative and homofermentative.
